B-cell chronic lymphocytic leukemia (CLL) is a type of cancer of the blood and bone marrow and the most common form of leukemia in adults. It's characterized by the accumulation of small, mature-appearing neoplastic lymphocytes in the blood, marrow, and secondary lymphoid tissues. The major signaling pathways involved in the tumorigenesis and in the resistance to the various anticancer therapies includes Notch, Hedgehog, and Wnt. Indeed, Notch signaling which regulates cell-fate determination during development and maintains adult tissue homeostasis was found recurrently mutated in CCL;about Wnt signaling, it also regulates crucial aspects of cell fate determination, cell migration, cell polarity, neural patterning and organogenesis during embryonic development and the Wnt/PCP pathway contributes to numerous aspects of CLL pathogenesis. Concerning Hedgehog, It exerts its biological effects through a signaling cascade that culminates in a change of balance between activator and repressor forms of glioma-associated oncogene (Gli) transcription factorsand its inappropriate activation occurs in several human cancers, including hematological neoplasms.
